The active compounds in Shiunko, shikonin and its derivatives were reported to be the accerleration of the proliferation of granulation tissue, anti-bacterial effect, anti-inflammatory effect and anti-tumor effect. A simple and rapid HPLC method was applied for simultaneous analysis of seven marker substances in Shiunko including shikonin, acetylshikonin, β,β-dimethylacrylshikonin, isobutrylshikonin, deoxyshikonin, isovalerylshikonin in Lithospermun erythrohizon and ferulic acid in Angelicae Radix. The HPLC method was carried out within 35 minutes by using Hypurity column (250 x 4.6 mm) and an isocratic solvent system of acetonitrile and acetic acid solution ( pH=2.56 ) . The marker substances in Lithospermun erythrohizon are unstable under light, oxygen, and heating. In this study, we used seven vegetable oils, including sesame oil, olive oil, soybean oil, safflower seed oil, peanut oil, sunflower seed oil and mineral oil to extract the bioactive substances in Lithospermun erythrohizon and Angelicae Radix, then compare the heat stability and photo stability of bioactive substances in different vegetables oils. The results showed that the bioactives substances in olive oil and mineral oil are more stable than others. In order to overcome the disadvantage the traditional ointment, we used mineral oil and olive oil to replace sesame oil and modified the product of Shiunko and then estimated percutaneous parameter of different products. The percutaneous absorption studies were performed using a modified Franz cell system through rat skin to assess the penetration of shikonin and its derivatives, and then compare the relationship between the molecular structure and permeability. The results demonstrated that only shikonin and acetylshikonin possessed the penetrated ability through the full thickness rat skin.
