The effects of androgen and estrogen are mediated through the AR and ERα, respectively. After ligand binding, heat-shock proteins dissociate from the receptors, and the AR or ERα translocate to the nucleus in which it regulates transcription by binding to discrete DNA sequences termed HREs in the promoter regions of target genes and interacting with other cofactors to regulated AR and ERα’s transactivity. In previous findings, we know the sex hormones induced AR or ERα-mediated gene regulation may involved in the formation of cancers, but the accurate mechanisms are still in vain. In this study we search the cofactor proteins for interacting with AR and ERα and identify their functions.
