Summary:碩士 === 大葉大學 === 分子生物科技學系碩士班 === 99 === In this study, we characterized two white spot syndrome virus (WSSV) structural proteins, VP15 (WSSV-T1 ORF269,GeneBank accession no. AF440570) and VP32 (WSSV-T1 ORF253,GeneBank accession no. AF440570). Temporal transcription analysis showed that VP15 is expressed in the early stage of WSSV infection. Yeast two-hybrid assays revealed that VP15 associated with VP19, VP26, VP95, VP51C, VP136, VP664, pmRACK1, F1ATP synthase beta subunit and itself. Yeast two-hybrid assays also demonstrated that VP32 interacted with VP187, VP19, VP60B, pmCBP, pmRACK1,F1ATP synthase beta subunit and pmCBP. Western blot analysis of viral protein fractions suggested that VP32 is a viral envelope protein. Membrane topology prediction showed that the VP32 is exposed outside of envelope without transmembrane domain.
