But soft rot bacteria in cultivation course seriously damages cutflower production. Most farmers can’t renew the aging and diseased rhizome bulbs due to the high cost of imported bulbs. It is necessary to establish an in vitro plant regeneration system of Z. aethiopica L. Nutrient requirement of plant is dependent on plant species. In the first stage of this study, shoot tip were cultured in MS basal medium (1/2, full, double strength of MS salts) combined with sucrose (3%, 6%). The result showed that full MS basal medium combined with 3% sucrose is optimal for shoot growth, after four weeks relative growth rate of shoot attained to 91%. In the second stage, shoot basal discs derived from in vitro plantlets are used to induce mutiple shoot with application of full MS basal medium combined 3% sucrose supplemented with 2,4-D (0、1 mg l-1) combined with TDZ (0、0.5、1 mg l-1) and BAP (0、0.5、1 mg l-1). The result showed that 0.5 or 1 mg l-1 TDZ can induce multiple shoot formation with the average number 3.17 and 3, irrespectively. The composition of 0.5 mg l-1 TDZ and 0.5 mg l-1 BAP can promote shoot growth with the average height 3.37 cm. The shoots were subcultured to MS medium for rooting.
