Now, Cyathea lepifera sick in Taiwan counties and cites, include Lanyu and Turtle Islands. Even less than the reproductive plants have also died and morbidity, but it has not tree stems (snake wood) who have no signs of morbidity. Cyathea lepifera that it’s to collect sporophyll specimens and set up SOP (Standard Operating Procedures) of spores framework. Four test for viability and maturity of the spore color, obtain optimum maturity of sporophyll. So Cyathea lepifera phenology and spore maturity phenomenon observed. At four conditions: room temperature, 4℃, –20℃ and –80℃ storage temperature, find the best suitable storage temperature, and prolong life of Cyathea lepifera spore viability, and set up spore bank. Best mature spores carried gametophyte sexual reproduction, and set up SOP (Standard Operating Procedures) of spore germination and gametophyte growth of Cyathea lepifera. Cyathea lepifera sporophyll that best collection period in late July to August to September. Select the ring with partial cracking of pinnate as a priority objective, best collected sporophyll maturity and receive the best quality spores. Best maturity spore is dark brown spores. Dark brown spores at –20 ℃ and –80 ℃ storage temperature, in 18 months of storage after, the average germination viability to 100%. At 4 ℃ storage temperature, 18 months of storage after, the average germination viability to 99%. Studies have shown spores deeper the color, the better spore viability. When the spore gets bigger and more full, its better viability, also suitable for long-term storage, delayed germination time when long-term storage. At 6 months after, immature spores stored at room temperature, that it has completely lost germination vitality. At 6 months after, dark brown spores stored at room temperature, that it has germination vitality to 48 %. Spore bank that it research shows and it needs Cyathea lepifera mother tree to add fresh and ripe spores every year. Otherwise, spore bank storage impact and affect the ability of natural regeneration and succession. Cyathea lepifera isn’t sexual reproduction illness, in spore germination and gametophyte growth of Cyathea lepifera. Plot developmental stages in growth and stabilize. In day/night temperature 30/25℃and sunlight at 12 hours one day. In the first weeks, spore germination starts with the rupture of the exine and formation of rhizoid and protonema cell. In the second weeks, the filamentous protonema divide and transit to two-dimensional prothallium. In the 6 weeks, heart-shaped, three-dimensional thalli with the apical meristem formation. In the 10 weeks, gametophyte with antheridium and archegonium formation. In the 12 weeks, the first sporophytic frond visible. In 12 months, the 10cm fern plant visible.
