The nanometer-scale inner cavity of the apoferritins can be filled with many kinds of inorgnic materials. In this study, we used the nano-biotemplates from horse spleen ferritin, which was immobilized on the self-assembled monolayer modified electrode, to synthesis zinc oxides in the inner cavity of the apoferritin. The photoelectroic properties of the ferritin immobilized electrode and the ferritin immobilized electrode with zinc oxides (ZnO) were characterized by cyclic voltammetry (CV), linear sweep voltammetry and chronoamperometry. To enhance the chances of proteins immobilized on the electrode, we used three kinds of thiols with different mixing ratios: 3,3’-dithiodipropionic acid di(N-hydroxysuccinimide ester (DTSP), 3-mercaptopropionic acid (MPA) and 3-mercapto-1-propanol (MPOH) to modify the electrodes. According the modified results, we chose the mixing ratios of the highest surface coverage to be our modified formula. The results showed that 1 mM DTSP + 10 mM MPA had the highest surface coverage, 0.627. The metal core inside the ferritin was removed by ethylenediaminetetraacetic acid using CV. Then, we used the chemical precipitation method to procedure ZnO. The results of CVs showed that the reduction peaks of zinc and zinc oxides occurred at -0.70 V and -1.12 V. We used the 1600 LUX of 460 nm blue light by LSV to decide the potential for light on-off experiments, -0.3 V. We found that the ZnO-ferritin electrode displayed a clear photocurrent under -0.3 V.
