In this study, we used a cell-lineage tracing approach to identify a population of ovarian surface epithelium (OSE), which expresses anti-Müllerian hormone type II receptor (AMHR2) in Amhr2-Cre/+;R26R-mTmG/+ reporter mice. We found that the Amhr2-Cre positive cells were distributed in a mosaic pattern throughout the OSE. By quantification of Amhr2-Cre+ derivatives in the OSE of Amhr2-Cre/+;R26R-mTmG/+ reporter mice, we showed that the GFP+ cells accounted for 30% of OSE at the age of 3 and 5 months. This observation indicates that AMHR2+ population in OSE is essentially stable through several rounds of natural ovulation in Amhr2-Cre/+;R26R-mTmG/+ reporter mice at the age of 3- and 5-month-old. Using BrdU pulse–chase method for 3-month trace, we found that the OSE of Amhr2-Cre positive lineage had more BrdU label-retaining cells (LRCs) in comparison with that of Amhr2-Cre negative lineage. Besides, using OSE colony forming assay, we found that Amhr2-Cre+ derived colonies could be passaged to second generation, indicating that Amhr2-Cre+ derivatives of OSE may contain a stem/progenitor population.
