Semi-solid detergent agar: an alternative for the conservation ex situ of Cladosporium spp. (Ascomycota: Cladosporiaceae)

The genus Cladosporium is ubiquitous and has a broad environmental representation. For the conservation of strains of this fungal genus, several methods of choice have been used, among which are cryopreservation and lyophilization. However, other alternative and complementary methods reduce conserva...

For the conservation of strains of this fungal genus, several methods of choice have been used, among which are cryopreservation and lyophilization. However, other alternative and complementary methods reduce conservation costs. The purpose of this investigation was to evaluate the feasibility of the conservation of 92 strains of Cladosporium in the medium Semisolid Detergent Agar (2 % bacteriological agar, 0.05 % Tween 80 and 100 ml of sterile distilled water). Viability, purity and morphological integrity were determined up to three years after initial conservation. For conservation, 1.5 ml Eppendorf™ tubes with 1 ml of medium were used, which were inoculated with spores obtained from a 7-day culture in Papa Dextrose Agar and kept at 4 °C. During the evaluated period, the strains maintained viability with values higher than 90 %, so this can be an alternative method for the conservation of strains of Cladosporium in the short or medium term.
