Summary:Objective: To investigate the role of miR-34a on the mice knee osteoarthritis. Methods: Mice knee osteoarthritis model was established, and HE staining was used to evaluate the Mankin score of knee cartilage injury. Then the chondrocyte of knee joint was cultured. RT- PCR was used to detect mRNA expression of miR-34a, and Western Blot was used to detect expression of bcl-2 and MMP-13. Results: In the mice knee osteoarthritis model, Mankin score increased, expression of miR-34a and MMP-13 increased, and expression of bcl-2 decreased. Conclusions: In mice knee osteoarthritis model, over-expression of miR-34a leads to the increased expression of MMP-13 and the decreased expression of bcl-2, and promotes osteoarthritis progression. This suggests that silencing miR-34a may become a new method for prevention and treatment of osteoarthritis.
