Summary:The efficient synthesis of stable isotope labeled D12-Malachite Green with CD3OD as the precursor by methylation, condensation and oxidation was presented. The yields of D12-Malachite Green was 50.1 % based on CD3OD consumed. The product was ideatified by HPLC, MS, and NMR to be the target compound. Its chemical purity and isotopic enrichment were higher than 99.0 atom%D. The result showed that the D12-Malachite Green could be used as internal standard in the field of food safety.
