Summary:Objective: To study the effect of puerarin on osteoblast proliferation in vitro and Wnt/β-catenin signaling pathway expression. Methods: Osteoblasts MC3T3-E1 were cultured and divided into control group, 10-6 mol/L PUE group, 10-7 mol/L PUE group and 10-8 mol/L PUE group, and after 24 h of treatment, mRNA expression of osteoblast marker genes, apoptosis genes and Wnt/β-catenin pathway genes were determined. Results: ALP, Runx2, OC, Wnt1, Wnt2, Wnt3a, Wnt10b and β-catenin mRNA expression in 10-6 mol/L PUE group, 10-7 mol/L PUE group and 10-8 mol/L PUE group were significantly higher than those in control group while caspase-3, caspase-8, caspase-9 and caspase-12 mRNA expression were significantly lower than those in control group; the higher the puerarin dose, the higher the ALP, Runx2, OC, Wnt1, Wnt2, Wnt3a, Wnt10b and β-catenin mRNA expression in cells while the lower the caspase-3, caspase-8, caspase-9 and caspase-12 mRNA expression in cells. Conclusion: Puerarin can effectively promote the proliferation of osteoblasts and the activation of Wnt/ β-catenin pathway.
