Previous issue date: 2014-07-31 === As algas marinhas vermelhas do g?nero gracilaria sintetizam polissacar?deos sulfatados (PS) bioativos. Por?m muitos destes PS ainda n?o foram devidamente avaliados, como ? caso dos PS sintetizados pela alga comest?vel Gracilaria birdiae. Estudos anteriores mostraram que galactanas sulfatadas dessa alga tem efeito antiinflamatorio. Neste trabalho uma galactana (GB) de G. birdiae foi obtida e avaliada por diferentes testes. GB apresentou atividade anticoagulante no teste de APTT. Gb n?o apresentou toxicidade para c?lulas normais (3T3), mas inibiu a sobreviv?ncia de c?lulas de adenocarcinama de colo de ?tero (HeLa) e human pancreatic carcinoma (Panc-1) em cerca de 80 % (1.5 mg/mL). GB n?o foi capaz de sequestrar o radical OH nem o radical superoxido. Contudo, apresentou atividade doadora de el?trons em dois testes diferentes e apresentou atividade quelante de ferro (70% a 1.0 mg/mL) e de cobre (70% a 0.5 mg/mL). A presen?a de GB promove uma maior forma??o de cristais de oxalato de c?lcio dihidratado de pequeno tamanho, que ? a forma menos agressiva, isso porque GB ? capaz de interagir com o cristal e estabilizar essa forma. Al?m disso, GB (2.0 mg/mL) n?o foi citot?xico para c?lulas renais humanas (HEK-293). Os dados obtidos n?s levam a propor que GB apresenta um grande potencial para o tratamento de urolit?ase. === Red marine algae of the genus Gracilaria synthesize sulfated polysaccharides (PS) bioactive. But
many of these PS were not properly assessed, as is the case of PS synthesized by edible seaweed
Gracilaria birdiae. Previous studies showed that sulfated galactans this alga has anti-inflammatory
effect. In this work, a galactan (GB) of G. birdiae was obtained and evaluated by different tests. GB
showed anticoagulant activity in APTT assay. GB showed no toxicity to normal cells (3T3), but
inhibited the survival of cells of adenocarcinoma of the cervix (HeLa) and human pancreatic cancer
(Panc-1) 80% (1.5 mg / ml). GB was not able to hijack the OH radical or the superoxide radical.
However, showed activity electron donor in two different tests and presented iron chelator activity
(70% and 1.0 mg / ml) and Copper (70% at 0.5 mg / ml). The presence of a higher GB promotes
formation of crystals of calcium oxalate dihydrate small size, which is less aggressive, because GB
is able to interact with and stabilize the crystal that form. Furthermore, GB (2.0 mg / mL) was not
cytotoxic to human renal cells (HEK-293). The data lead us to propose that GB has a great potential
for the treatment of urolithiasis
