Foram selecionadas crianças de 6-15 anos, sendo 18 com SD e 23 não sindrômicas (controle). Inicialmente foram avaliados os índices CPO-D/ceo-d e o índice gengival (IG), realizada a coleta da saliva estimulada e determinados fluxo salivar (mL/min), pH e capacidade tampão. A contagem de Streptococcus mutans e Lactobacillus spp na saliva foi realizada após cultivo em meios seletivos e a quantificação de Porphyromonas gingivalis, Tannerella forsythia e Aggregatibacter actinomycetemcomitans, pela Reação em Cadeia da Polimerase em Tempo Real (qPCR). A dosagem de proteína total (PT), a atividade da Superóxido Dismutase (SOD) e da Glutationa Peroxidase (GPx) e a capacidade antioxidante total da saliva (TAOC) foram determinadas por espectroscopia, e o nível de Malondialdeído (MDA) por cromatografia líquida de alta performance (HPLC). Os resultados foram submetidos ao teste de Mann-Whitney e Correlação de Spearman (α=0,05). Não houve diferença nos índices CPO-D/ceo-d e IG entre os grupos. O fluxo salivar foi menor no grupo SD (p<0,0001). Não houve diferença entre os grupos com relação a quantidade de bactérias cariogênicas e periodontopatogênicas. Houve correlação positiva entre contagem de Streptococcus mutans e Lactobacillus spp e os índices CPO-D/ceo-d (r=0,621 e r=0,540, respectivamente), apenas para o grupo SD. A GPx e a TAOC não apresentaram diferença significativa entre os grupos, enquanto a dosagem de PT (p<0,0001), a SOD (p=0,0002) e o MDA (p=0,0016) foram maiores na saliva do grupo SD. Houve correlação positiva entre PT e IG (r=0,491), e negativa entre SOD e fluxo salivar (r=- 0,475) para o grupo SD. Pode-se concluir que: (1) O fluxo salivar reduzido nas crianças com SD... (Resumo completo, clicar acesso eletrônico abaixo) === Abstract The aim of this study was to evaluate salivary factors related to dental caries and periodontal disease in children with Down syndrome (DS). Forty-one children aged 6- 15 years, being 18 DS and 23 non-syndromic (control) were selected. First, DMFT/dmft and gingival index (IG) were evaluated and stimulated whole saliva was collected. Then, salivary flow (mL/min) was calculated and pH and salivary buffering capacity were measured. Streptococcus mutans and Lactobacillus spp counts were determined after culture on selective medium and Real-time Polymerase Chain Reaction (qPCR) was used for absolute quantification of Porphyromonas gingivalis, Tannerella forsythia and Aggregatibacter actinomycetemcomitans. Total protein dosage (TP) dosage, superoxide dismutase activity (SOD), glutathione peroxidase (GPx) and total antioxidant capacity of saliva (TAOC) were quantified by spectroscopy. Malondialdehyde level (MDA) was quantified by High-performance liquid chromatography (HPLC). Data were analyzed using Mann-Whitney test and Spearman correlation (α = 0.05). There was no difference in DMFT/dmft and GI index between groups. Salivary flow was significantly lower in DS group (p <0.0001). Related to cariogenic and periodontopathogenic bacteria there was no statistically significant diference between groups. A positive correlation between counts of Streptococcus mutans and Lactobacillus spp and the DMFT/dmft index (r=0,621 e r=0,540, respectivelly), was verified only for the DS group. No significant difference between groups was observed for GPx and TAOC, while TP (p<0.0001), SOD (p=0.0002) and MDA (p=0.0016) were higher in saliva of DS group. There was a positive correlation between TP and GI (r=0,4915), and a negative correlation between SOD and salivary flow (r=-0,4751) in DS group.
