There exist two important single point mutations on the FY gene, which results in different Duffy genotypes and consequently varied susceptibility to vivax malaria. Our study has a two-pronged objective. Firstly, using an erythrocyte-binding assay, we show that rPvDBPII binds to the red cells in a concentration dependent manner. We observe, variation in the percent binding between donor erythrocytes of the same Duffy genotype despite similar Fy6 antigen expression. This could suggest the presence of other red blood cell polymorphisms surrounding the Fy antigen or post-translational modifications. Secondly, we see that, rPvDBPII binds to a chymotrypsin sensitive unknown receptor on Duffy negative erythrocytes.
