Most of the current studies focus on the detrimental effects and adverse consequences of inflammation whereas the beneficial effects of inflammation are largely neglected and poorly exploited. Previous literature suggests that the factors that determine the trade-off of the beneficial versus detrimental effects of inflammation involve regulation of immune-to-brain signaling and diverse functions of inflammatory mediators. Therefore, this dissertation investigated these opposing aspects of inflammation. In the first half of the dissertation (chapter 2 and chapter 3) we reported the discovery of euflammation, a type of inflammation induced under muted immune-to-brain signaling. Study of euflammation demonstrates that beneficial effects of inflammation can be elicited through modulation of neuroimmune communication. In the second half of the dissertation (chapter 4 and chapter 5), we created a genetic mouse model to study the diverse functions of an inflammatory cytokine, interleukin 1. Using this model we mapped out the distribution of IL-1R1 in the brain and revealed more cell type specific IL-1R1 mediated central IL-1 effects. Taken together, these findings havepartially unraveled the complexity of inflammation and provided therapeutic values for treatment of excessive neuroinflammation. 2017-10-25 English text The Ohio State University / OhioLINK http://rave.ohiolink.edu/etdc/view?acc_num=osu1496016916912326 http://rave.ohiolink.edu/etdc/view?acc_num=osu1496016916912326 unrestricted This thesis or dissertation is protected by copyright: all rights reserved.
