The goal of this research was to explore the potential of RNase U2 to improve sequence coverage in RNA modification mapping. Mutagenesis of recognition-site residues indicate that work cooperatively to bind ribonucleotides. Glu62, previously thought as a catalytic residue also proved to be more important in binding versus catalysis.
