Fillamentous bacteriophage Xf from Xanthomonas oryzae was quite stable in various conditions. It could be stored in Potato Sugar medium, phosphate buffer or distilled water at 40C for four months without affecting its infectivity. It could also be detected after having aprayed on the leaves of rice plants or in the field water for one month.
There were 17 sites of attachment for each host bacterium. It is much larger than that of filamentous bacteriophage of Escherichia coli.
Variety of ions on the solution exert different influsence on adsorption rate. The adsorption rates in ions used in this investigation were in the order: NH4+>Mg++>Na+>Ca++>H2O. The adsorption did not affected by the treatment of host bacteria with chloramphenicol. Although chloramphenicol stopped the growth of host bacterium, the adsorption was normal. This implied that the adsorption was not dependent on prtein synthesis, but on the native state of the bacterial cell walls.
Heat-killed bacterial cell did not adsorb any infective phage Xf. It appeared that heat treatment destoryed the structure of adsorption sites. This property was completely different from tadpole-shaped bacteriophages.
