Cholesterol and cholest-4-en-3-one showed significatantly peak tailing on C8 olumn, but not on CN column.   The retention times of cholesterol and cholest-4-en-3-one were related with pressure and temperature. The reteniton time of cholesterol was shorter than cholest-4-en-3-one on CN column, however longer on C8 column. In the range of the column, however longer on C8 column. In the range of the concentraction from 0.2∼20 mg/mL, the correlation coeffecient for the calibration of choleterol in 210 nm was 0.99985, and that of cholest-4-en-3one in 240 nm was 0.99998.   On CN column, cholesterol, cholesteryl chloroacetate, cholesteryl phenylacetate, and cholest-4-en-3-one were competely separated by keeping the pressure at 15 MPa and temperature programming from 80℃ to 60℃ at the rate of-3℃ per minute.   After improving the hardware of the system, the on-line SFE-cholesterol oxidase-SFC system was found efficient in cholesterol analysis. Cholest-4-en-3-one could be detected in 10 minutes after the cholesterol started reacting with enzyme. The correlation coefficient of the calibration curve was 0.9974. 洪良邦 1995 學位論文 ; thesis 80 zh-TW
