Summary:碩士 === 中山醫學院 === 生物化學研究所 === 84 === The prevalence of asthma in scool-age children in Taipei increased from 1.3% to 10.8% during the past twenty years .It is found that HDM (house dust mite ) is tje most important allergens in Taiwan ,especially for asthmatic children.this study is divided into two parts.In the first part the author used the affinity puriffication of human sera that attaches specific Der p5 antibodies withthe Der p5 recombinant fusion protein .The result will elute antibody in mite extraction and rDer p5 fusion protein to corresponding to components of Mr 14kd and 42 kd that can identify the natural allergen -Der p5 .The samplesare drawn from the 12 families in Taichung .Inclusion criteria:the family must has a child attacked asthma .The dust samples were collected from the mattress and the floors of thee bedroom and living room of the children homeenviroment from Oct 1994 to Sep 1995 .In the research finding ,the highest concentration of dust samples ranges from 6-8000 ng/g fine dust ,The highest samples were oberved the mattress .The lowest concentration in the floors of the bedroom.In the second part, the study is focused on the reaction frequency and relationsship betwwen antibodies(IgG and IgE)and purified mite allergen (Der p5) in different groups ,according to the result ,the Der p5 has the significance in the clinical finding .THe antibody (IgE) in the serum has the highly affinity and frequency in the children of asthma
