Simultaneously exposured to MC540 and white light inactivating enveloped viruses. Furthermore, the prelight-activated MC540 (pMC540) has been shown in earlier studies to be effective against certain types of tumor cells and viruses. Hepatitis B is a public health problem of worldwide importance. Hepatitis B virus (HBV) infection causes acute hepatitis and a chronic carrier state. In the present study, the primary culture of DHBV positive ducklings' hepatocyte system and the DHBV chronic carrier duck as the in vivo system were employed for investigat- ing the feasibility and effectiveness of MC540, as a photosensi- tizer for inactivating duck hepatitis B virus (DHBV). Addition of MC540 to the cultured cells, but without light irra- diation had an about 50% inactivation rate of intracellular DHBV and no influence on the extracellular release rate of DHBV. With light irradiation, when the dosage of MC540 was 140ug/ml, the inactivation of intracellular DHBV was almost complete, and the release rate in the medium was lowered to about 1% of the con- trol group. The prelight- activated MC540 at a dosage of 140ug/ml could have the corresponding effectiveness of light irradiation. The anti-DHBV activity of MC540 was also studied by the in vivo system of DHBV chronic carrier ducks. The result revealed that, within the administration of MC540 period, the concentration of the DHBV DNA in the serum of group pMC540 and group nonactivated -MC540, decreased to 36.6% and 44.8% of preadministration value, respectively. Moreover, the biochemical values in the serum of experimental ducks were found almost no significant changes during MC540 administration period.
