Results revealed that Lu-zhu strain has the highest resistance level to mevinphos, and Wu-fengstrain has the highest resistance level to methamidophos. In 2-D gel electrophoresis analysis, we found there were many differences in protein constitution between resistant strains and susceptibleLi-shan strain:Proteins of pI 6.22,MW 19.5 KD;pI 6.40,MW 22.0;pI 6.50,MW 18.0;pI 6.55,MW 23.5; pI 6.60,MW 25.0;pI6.65,MW 31.0;pI 6.85,MW 23.0;pI 7.45,MW 17.0 KD in Lu-zhu strain, and pI5.10,MW 18.0;pI 5.30,MW 28.0;pI 5.91,MW 31.0 KD;pI 5.95,MW 34.0;pI 5.96,MW 31.5 KD;pI 6.00,MW 26.0 KD;pI 6.01,MW 19.0 KD;pI 6.15,MW17.0 KD;pI 6.53,MW 16.5 KD;pI 6.58,MW 15.5 KD;pI 6.61,MW 17.5 KD;pI6.73,MW 25.0 KD;pI 6.90,MW 15.0 KD;pI 7.03,MW 16.0 KD in Wu-feng strain weredifferent from Li-shan strain. In the dot assay, among the 52 DNA immobilized on nylon membrane, the firststrand cDNA made from the mRNA of all three strains were hybridized to GST ,mdr3, and cytochrome P450A1. This might indicate other genes could be involved in the resistance of the insects to organophosphorous insecticide. This mightexplain why the resistance of the diamondback moth to insecticides is common and there could have other pathway could be involved in detoxification.
