In liver microsome from male Wistar rat, TRA transformed into two products, MA1 and MA2 in the presence of NADPH. But, in liver microsome from female Wistar rat, TRA transformed into only one product, designed as MA1. The sex difference was observed in the formation of MA1 and MA2 by the hepatic microsome isolated from Wistar rat. Castrated male Wistar rat and ovarectomized female Wistar rat decreased MA1 produced and then administration of estradiol recovered MA1 product. MA2 can not found in the castrated male Wistar rat and MA2 was produced in administration of testosterone to castrated male Wistar rat and ovarectomized female Wistar rat. From above studies suggested that testosterone and estradiol were involved in the MA1 formation and MA1 to MA2 was testosterone dependent. Both steps of reactions were enhanced by pretreatment of Wistar rat with phenobarbital.
