anti-GAD65 (anti-glutamic acid decarboxylase), one of the
major autoantibodies that exist in insulin-dependent diabetes
mellitus(IDDM), and to analyse the relationship between HLA
class II molecules and anti-GAD65. In this study we established
96-well format immunoprecipitation radioligand method to detect
anti-GAD65 antibody, and established SSP-PCR(sequence-specific
primer-PCR) method for typing HLA class II gene.
The study population consisted of 167 IDDM patients, 125
relatives of IDDM(parents and siblings), and 105 healthy
control. The results showed that:(1)The prevalence
of anti-GAD65 in Taiwanese IDDM was 58.33﹪. The prevalence of
anti-GAD65 decreased with the increase of the duration of
disease, and at the same time, the titer of anti-GAD65 declined
with the increase of the duration of disease.(2) Two of the HLA
haplotypes:DRB1*17-DQA1*0501-DQB1*0201,DRB1*04-DQA1*0302-
DQB1*0401 showed strong association with IDDM(p<0.05). However,
three of the HLA haplotype :DRB1*11-DQA1*0501-DQB1*0301,DRB1*12-
DQA1*0601-DQB1*0301,DRB1*14-DQA1*0101- DQB1*0503 were protective
against IDDM(p<0.05).(3)HLA genotypes, DRB1*09/09 and
DQB1*0303/0303 were associated with an increase in the frequency
of anti-GAD65(p<0.05). Nevertheless, DRB1*04-DQA1*0302-
DQB1*0401 could be correlated wition will be necessary.(5)The
HLA haplotype: DRB1*17-DQA1*0501-DQB1*0201 had association with
early age of onset.(6)The DQα1( Arg52)molecules had larger
effect than DQβ1(Asp57)molecules to susceptibility of IDDM.
In this study, we established 96-well format immunoprecipitation
radioligand method to detect anti-GAD65 antibody. It's the first
time to analyse the prevalence of anti-GAD65 in Taiwanese IDDM,
and to analyse the relationship between the prevalence and
titer of anti-GAD65 with HLA class II molecules.
