Summary:碩士 === 中國醫藥學院 === 中國醫藥研究所 === 86 === San-Huang-Hsieh-Hsin-Tang ( SHHHT ) , an traditional formula in China , consis ts of three crude drugs : Rhei Rhizoma , Coptidis Rhizoma and Scutellariae Rad ix., are widely used in the treatment of hypertension , anxiety , gastritis an d several hemorrhage disorders. In this study , the pharmacokinetics and bioav ail- ability ofthe SHHHT in rabbits were evaluated.A method for simultaneous d etermination of the active constituents of the SHHHT, included baicalin , berb erine and emodin in plasma by self-developed HPLC assay was described.After IV administration of the SHHHT , ( contains baicalin 6.488 mg , berberine 5.506 mg and emodin 0.250 mg ) to the rabbits the plasma level-time courses of these three compounds could be adequately described by an open two-compartment mode l. The elimination half-lives of baicalin , berberine and emodin were 145 , 90 and 57 minutes respectively.The absolute bioavailability of these three compo unds after oral administration of the SHHHT were 21.17 , 6.54 and 2.79 % for b aicalin , berberine and emodin respectively. This result suggest that repeated dose is necessary for SHHHT in clinical use.
