Both pertussis toxin (PT) and filamentous hemagglutinin (FHA) have been used as the components of acellular vaccine for pertussis infection. Several studies suggested that PT can induce a very strong proliferative response for human lymphocytes and to lesser extent, murine lymphocytes. To further characterize the cellular immune response, intracellular staining of DNA with DNA dye and anti-BrdU antibody, antigen-reactive T cell frequency by limiting dilution and a-MHC-II mAb blocking effect of peripheral blood mononuclear cells (PBMCs) were analyzed. The data suggested both the proliferative response and antigen-reactive cell frequency of PT stimulated PBMCs were similar to that of Con A stimulation, which was much higher compared to that of FHA stimulation. In addition, PT can also induce a good cellular proliferative response of cord blood mononuclear cells which are regarded as a population of naive cells. With further characterization of PT-specific cellular immune response might shed light into further design of pestussis vaccine and application of PT as an immunological adjuvant.
