Therefore, the purpose of this study is to investigate the effect of different levels dietary protein on the geneexpression of amyloid precusor protein (APP695) gene. Twenty-four male, 8 wks old SD rats were randomly assigned into groups of three according to their body weight and fed diets containing 8% protein (low protein group),18%protein (control group) and 36% protein (high protein group), respectively. Feed intake and body weight gain were recorded daily. Seven and 11d after feeding the rats with experimentaldiets, the cerebral cortex of each rat was excised and total protein and total RNAwere extracted. The amount of total RNA from different samles were analyzed by RT-PCRtechnique and slot blot to study the effect of dietary protein on APP695 expression at transcriptional level. Western blot analysis were used to detect the change of change of the amount of APP695 by dietary protein. After 7 days, the wet weight of liver and kidneys was significantly increased, hoeever intake tended to be supressed by high protein feeding to the rats. At transcriptionallevel, APP695 mRNA was not affected by the regimen. However, the amount of brain APP695was decreased significantly by feeding the rats with low protein diet. After 11 days, feedintake was decreased and liver kidney weights were increased by high protein feeding.The amount of APP695 mRNA was not affevted by dietary protein. However, the amount of APP695 tended to be increased as dietary protein increased in the diets.The results of this study suggest that APP695 gene expression in rats brain can bealtered by dietary protein at translational level.
