However, having the tendency to become unstable upon the application of heat, they are difficult to determine by GC and other techniques. Although the active components of rhubarbs can be separated by HPLC and detected by photodiode array detector, their complexity and low content make identification by photodiode array detector or conventional UV difficult.In this thesis, the use of LC/ES/MS to analyze these components was investigated. An analytic method is developed to separate and identify the components of rhubarbs by using C18 column and gradient elution to first separate the components that are then determined by ES/MS. In addition, the effect CapEx, mobile phase composition, mobile phase additives, and flow rate have on ES/MS detection as well as the linearity of LC/ES/MS and detection limit are evaluated.
