Description
Summary:碩士 === 國立雲林科技大學 === 環境與安全工程技術研究所 === 86 === This research focus on the isolation of microbes which could degradate the aromtic compounds from the coast of Taiwan. One phenol utilizing bacteria, The bacteria is pseudomonas cepacia, Ph-2 after determine by API20NE system. Using the shaking method, the optimal cultivation condition for pseudomonas cepacia was conducted. From the research, the culture condition of pseudomonas cepacia : 0.1%(wt./vol.) phenol as sole carbon source ; nitrogen 1.0 g/L ; phosphorus 0.25 g/L, the growth condition : pH 7;temperature 30℃. The average specific growth rate 0.0339 hr-1.Pseudomonas cepacia Ph-2 were cultivated in the fermentor under optimal condition. When cell cultivation to the log phase or stationary phase, and the fermentation broth blended the carriers . such as soybean mill, wheat brans. Then dried using freeze-dry and hot air-dry to produce bioaugmentation agent, the latter method is better than former. The latter posses a viable count of 1.9×1013 CFU/gram product. The capability of phenol biodegradation was evaluated by shaking cultivation method at room temperature. After 48 hours treating, phenol biodegradation efficiecy are 99.8% by hot air-dry product, and it need 60 hours for the freeze-dry agents. We can conclude that pseudomonas cepacia Ph-2 can be make as bioaugmmentation product which effective to phenol degradation. In the furture , we can further apply to marine oil-spill treatment.
