A total 0f 36 male Sprague-Dawley (SD) rats were selected for study. They were divided into 6 groups of 6 rats as follows: 1)Normal control group: received intracortical injection of 8μl PBS solution ; 2)Control group: received intracortical injection of 8μl 100mM FeCl3 solution;3)VA2 group: received intraperitoneal injection(i.p.) of VA 200mg/kg 30 min prior to FeCl3 administ-ration ; 4) VA1 group: received VA 100mg/kg i.p. 30 min prior to FeCl3 administration ; 5)Contrast group:received phenytoin 10mg/kg i.p. 30 min prior to FeCl3 administration ; 6)Control contrast group:received PBS i.p. 30 min prior to FeCl3 administration. Epileptic seizures were verified by behavioral observations, and electroencephalography (EEG) and electromyo-graphy (EMG) recordings. Behavior , EEG and EMG were monitored from 1 hour to 5 hours after FeCl3 administration . The number of wet dog shakes (WDS) were counted at 10 min intervals throughout the experimental course. All 36 SD rats were used to measured by lipid peroxide levels in the brain and Luminol-chemiluminescence (CL) counts and Lucigenin-CL counts in peripheral whole blood 5 hours after FeCl3 administration. Our results indicated that VA 200mg/kg ,100mg/kg and phenytoin 10mg/kg decreased the incidence of FeCl3-induced wet dog shakes , and also decreased FeCl3-induced lipid peroxide level in the cerebral cortex. VA 200mg/kg also reduced Luminol-CL and Lucigenin-CL counts in peripheral whole blood which were evoked by Ferric chloride. Our conclusion is that VA 200mg/kg ,VA 100mg/kg and phenytoin 10mg/kg may decrease the incidence of FeCl3-induced wet dog shakes in rats, therefore , it has an anticonvulsive effect. VA also decrease the FeCl3 induced-lipid peroxide, and Luminol-CL and Lucigenin-CL counts increase , suggesting that VA has a suppressive effect on lipid peroxidation and free radical scavenging activities, suggestive anticonvulsive effect of VA results from its oxygen free radical scavenging activity , because previous studies has known that oxygen free radical plays an important role in FeCl3-induced epileptic seizures . Anticonvulsive effect of GE ,at least in part, results from its component of VA.
