This pathogenicity island controls the ability of bacterium to elicit the hypersensitive response (HR) in nonhosts. The hrpC operon is comprised of hrpF, hrpG, hrcC, hrpT and hrpV genes. HrpV is a negative regulator (Preston et al., 1998). HrcC is an outer membrane protein and helps the elicitor-like HrpZ travelling across outer membrane (Deng and Huang, 1999). The transcriptional start site and the activity of promoter of hrpC operon were confirmed by using primer extension and lacZ reporter gene system. The expressions of hrcC, hrcQb and hrpZ genes in the hrpF mutant were analyzed by western blotting with antisera against HrcC, HrcQb and HrpZ . Respectively the results suggest that HrpF positively regulate the expression of hrcQb and hrpZ genes. The results of cesium chloride density gradient suggest that HrpJ, HrcR and HrpT , but not HrpF protein would involve in structure protein include HrcC protein completely targeting to membrane.
