They also have certain relationships with the growth or inhibition of cancer cells. Interleukin-6 (IL-6) and interferon-γ(IFN-γ) are both cytokines, IL-6 can induce terminal differentiation on several cancer cell lines resulting in losing their ability to proliferate continuously then achieve the anti-tumor effect, on the other hand, IFN-γ has been proved owning the direct cytostatic effect on cancer cells. But, their relationships with oral cancer cells were rarely investigated. The aims of this experimentation were to study the effects of IL-6 and IFN-γ on the cell growth and the prostaglandin synthesis of human oral cancer cell lines and normal fibroblasts cell lines. In our laboratory, we established one human oral epidermoid carcinoma cell line from Chinese patient (OEC-M1), obtained another Caucasian oral epidermoid carcinoma cell line from ATCC (CCL-17) and obtained one normal human oral mucosa fibroblasts cell line from South Africa (CT'5). First of all, we observed the morphological changes of different cell lines with phase contrast microscope after addition of these two cytokines, and the cell growth curves were determined subsequently. Finally, the activity of prostaglandin endoperoxide synthase was measured using thin-layer chromatography and liquid scintillation spectroscopy. The results showed that when oral cancer cells treated with different concentrations of IL-6 and IFN-γ(50 U/ml, 100 U/ml, 200 U/ml), the cell growth of OEC-M1 cell line was promoted, and the cell growth of CCL-17 cell line had no significant effect when treated with IL-6 but was inhibited by IFN-γ. Both cytokines had no significant effects on the normal human oral mucosa fibroblasts cell line (CT'5). There were no significant morphological changes of these cell lines by different additions. In different situations, the production of prostaglandin E2 (PGE2), which representing the activity of PG synthase, had significant differences. When IL-6 was added to OEC-M1 cell line, PGE2 production amount was significantly increased in 200 U/ml group. When IL-6 was added to CCL-17 cell line, there are significantly increased PGE2 production amount in both 50 U/ml and 100 U/ml groups, especially in the 50 U/ml group. But in the 200 U/ml group, it's production amount was significantly decreased. And when IL-6 was added to CT'5 cell line, the production of PGE2 was significantly increased in all experimental groups, especially in the 200 U/ml group. The 50 U/ml group rank second, then the 100U/ml group. When IFN-γ was added to OEC-M1 cell line, the production of PGE2 was significantly increased in 50 U/ml group, but decreased in 100 U/ml group. And when IFN-γ was added to CCL-17 cell line, the PGE2 production amount was significantly increased in 50 U/ml group and 200 U/ml group. When IFN-γ was added to CT'5 cell line, the PGE2 production amount was also significantly increased in 50 U/ml group and 200 U/ml group.The results suggest that the effect of IL-6 and IFN-γon the growth of oral cancer cell lines may be modulated by endogenous prostaglandin synthesis.
