Extracts was purified by stepwise elution on a HyperD and a Prepacked Q column in 20 mMTris-HCl to 20 mMTris 1M NaCl (pH7.9). Successively four fractions called DHN, DHB1, DHB2, DHB3 were separated by Hyper D media. The active ingredient showed on the bound protein fraction DHB1 toward HB4C5. Using the same method purified the DHB1fraction by Prepacked Q column found active ingredient on bound protein fraction QHB2. The QHB2 fraction shown both growth promoting and immune stimulating more effective than Extracts and DHB1 toward HB4C5. Hydrolysate of muscle from freshwater clam shown efficiently on proliferation toward HB4C5 by protease enzyme. The optimal time and enzyme amount were 2 hrs and 2.36% under clam muscle 100g. Subsequent study on efficiency of Hydrolysate by different enzyme, which were Alcalase, Amyloglucosidase, amylase, Flavourzyme, Neutrase, Protamex, and protease. Hydrolysate were hydrolyzed by Protamex and protease show cell proliferation, respectively at the optimal time and enzyme amount were 3 hrs and 8.88% under clam muscle 400g. Meanwhile hydrolysate by protease stimulated IgM production approximately 2-3 fold by UF (YM100 、YM30). Zwe-Ling Kong 龔瑞林 1999 學位論文 ; thesis 77 zh-TW |
