Summary:碩士 === 中國文化大學 === 應用化學研究所 === 87 === Cancer has been the leading cause of death in Taiwan Kinma area since 1984 1 .It is estimated that more than one Fifth population in Taiwan Kinma area will die of cancer.Among The causes of cancer death in Taiwan-Kinma area,lung cancer Is the first leading death cause for female,the second leading death cause for male,and the first for all In 1997,totally 5857 men and women died of lung cancer.obviously,lung cancer is a major life-threatening health problem in Taiwan. Two types of lung cancers make up 95% of all primary lung neoplasms 2.They are small cell and non- small cell lung carcinomas (SCLC and NSCLC).In Taiwan,SCLC omprises 12-15% and NSCLC comprises 85-88%.NSCLC includes squamous cell carcinomas,adenocarcinomas,and large cell carcinomas.SCLC has better response rate to radiotherapy 80-90% and chemotherapy 90%,compared to 30-50% and 30-40%,respectively,in NSCLC. However,the overall 5-year survival rates of SCLC and NSCLC are less than10%,indicating the highly malignant nature of lung cancers.In recent years,many oncogene and tumor suppressor gene products,such as c-Myc3,4 and p53 5,were shown to be modified be modified by O-linked N-acetylglucosamine (O-1cNAc). In this study we will use molecular-pathological Methods to investigate the functional and topological Regulation of glycosylation-related genes in the biology and clinical behavior of lung cancer,with emphasis on tumor invasion and metastasis.Lung carcers have been shown to have a variety of genetic abnormalities,which include deletions,amplifications ,activation of oncogenes (RAS,MYC,c-erbB-2,and BCL-2),and inactivation of tumor suppressor genes(RB, P53, and P16 INK4A) 6 .The genetic abnormalities of lung cancers were vigorously studied in recent years 7-10 however,recently no single genetic marker is found to be sensitive and specific for efficient diagnosis or therapy,indicating the multifactorial nature of lung cancer tumo-rigenesis and the necessity of multivectorial approach to lung cancer research.Molecular pathology is the implementation of Molecular biology technology to pathological sciences,combining the beauty of biochemical analysis and in situ morphological observation. It is under rapid development and be applied to almost any field of disease-oriented biomedical research.In this study,we will pursue molecular pathology approaches to the study of lung cancer as well as the clinical pathological methodology of lung cancer research.
