Summary:碩士 === 長庚大學 === 基礎醫學研究所 === 88 === Gas7, a growth-arrest-specific gene isolated from serum starved mouse fibroblast cells, is expressed in the brain and is required for neurite formation in cultured mouse cerebellar neurons (Ju et al., 1998). Here, I report that Gas7 is involved in morphological differentiation in PC12 pre-neuronal rat pheochromocytoma cells. I inhibited the endogenous rat Gas7's production by antisense nucleotide complimentary to the translation initiation region of rat Gas7 mRNA and found that it would reduce the number and length of NGF-promoted neurite outgrowth, but did not involve in the cell cycle. Gas7 interacting protein (Gas7 N-ip) gene was isolated by yeast two hybrid system. It, just like Gas7, is expressed in the brain. I had prepared the antibody that there was good Sensitivity and specificity for detecting Gas7 N-ip, and found that its molecular weight is near 60 kDa.
