Summary:碩士 === 中山醫學院 === 生物化學研究所 === 88 === Increasing evidence of free-radical generating agents and inflammatory process suggests accumulation of reactive oxygen species involving hepatotoxicity.In this study,we use tert-butylhydroperoxide(t-BHP) tto induce hepatotoxicity in vitro and in vivo to assess the antioxidative bioactivity of color extracts of dried Hibiscus sabdariffa flowers(HS-colors).The investigations showed that pretreated-HS-colors(0.05-0.10mg/ml) significantly decreased the leakage of lactate dehydrogenase(LDH)''alanine aminotransaminase(ALT)''aspartatee aminotransferase(AST) and the formation of mmalondialdehyde(MDA) induced by 30 min treatment with t-BHP in primary cultured rat hepatocytes. In vivo study(rats), histopatthological evaluation of rat liver showed that groups treated with HS-colors at concentration of 50-200mg/kg for 5 day orally then with single dose of t-BHP(ip;0.2mmol/kg) reduced the incidence of liver lesion including inflammatory leukocytes infiltration and necrosis. Furthermore, groups pretreatted HS-colors at concentration of 50-200mg/kg for 5 day orally decerased the serum index(LDH''ALT''AST)of hepatic function and liver lipid peroxidation induced by t-BHP. In addition to scavenging free radical property,HS-colors might exert its protective effect through modulating intracellular detoxication enzyme such as catalase.
