Detection of organophosphorous pesticides residues based on the inhibition of immobilized acetylcholinesterase using an amperometric biosensor in a continuous flow system

碩士 === 國立東華大學 === 化學研究所 === 88 === A carbon paste electrode modified with choline oxidase (ChOx) and horseradish peroxidase (HRP) was designed as the amperometric biosensor for organophosphorous pesticides (OPPs) determination. The OPPs was measured based on the inhibition of immobilized acetylcholi...

The OPPs was measured based on the inhibition of immobilized acetylcholinesterase (AChE) as a reactor mounted before the paste electrode in the continuous flow system. Two different sources of AChE from eel and house fly head were studied respectively. The better detection limit for paraoxon (10ppb) and carbofiran (23ppb) was found in the latter AChE reactor with a linear range of 8-100ppb. Two different type of AChE reactors (CPG and Nylon-6 tubing) were studied also. The better detection limit for paraoxon (63ppb) was found in the latter AChE reactor with a linear range of 50-300ppb. A new design was developed to prevent the adverse effect of ChOx during AChE reactivation process. This system is fully automatic and the throughput of 5 sample h-1 is obtained with a good reproducibility. The correlated data from spiked river water and fruit juice real sample shows the feasibility of this system for environmental application. Hun-Chi Shu 蘇宏基 2000 學位論文 ; thesis 0 zh-TW
