A polyclonal morphine antibody immobilized on the matrix of controlled porous glass (CPG) as reactor was mounted in SIA system to on-line affinity extraction the morphine in the urine sample during analysis. The deuterated analogue (morphine-D6, 100ppb) as internal standard was used for quantitative purpose. The 10 ppb and 50 ppb limit of detection (S/N>3) were obtained in SIA-APCI-MS and SIA-ESI-MS, respectively. It shows at least six times sensitive than 300 ppb (the official cut-off of morphine in the urine). A good specificity was found that expect heroin and codeine have extremely slight interfering (no effect in normal analysis), most abused drugs including the morphine-glucuronide have no obviously interference. The time for one analysis is only 13 min and no significantly activity decline during 6 weeks testing shows a acceptable stability for measurements. The results obtained from the real urine sample measuring by GC-MS and this method shows good accordance.
