Description
Summary:碩士 === 國立臺灣大學 === 微生物學研究所 === 88 === During Mar. 1999- Mar.2000, wild rodents were captured for the examination of helminthic parasites and Babesia microti. A total of 71 animals, including 38 Rattus coxingia, 18 Bandicota nemorivaga, 10 R. losea, 2 Mus formosanus, 2 R. norvegicus, and 1R.rattus, were collected fromYang-ming-shan, Bei-tou and San-xia in Taipei, Yen-ping in Taitung, Qiao-tou and Niao-song in Kaoshung. Hlminthes wre preserved in 70 % ethanol or 10 % formalin, or stained and mounted. The overall infection rate was 97 %. Twenty helminth species, including 13 nematodes, 2 trematodes, and 5 cestodes, were found. Monningia monningia, Nematospira sp., Platynosomoides sp., Ascaris sp., and Mesocestoides sp. were recorded for the first time from rodents in Taiwan. Angiostrongylus cantonensis, Capillaria hepatica, Syphacia obvelata, Hymenolepis nana, H. diminuta, Raillietina sp. were zoogenic parasites between human and murine. H. diminuta was the most commonly seen (50.7 %)parasite in rodents. In addition, Nipponstrongylus braziliensis、Strongyloides ratti、Trichuris muris、Capillaria sp. 、Protospirura sp. 、Platynosomoides muris、Rictularia sp. 、Heterakis sp. 、Cysticercus fasciolaris were also found in rodents. Comparison on prevalence rates obtained by examination of Giemsa-staind blood smear and PCR amplification of B. microti DNA were made. Prevalence rate of B. microti infection among the 63 rodent blood specimens obtained by microscopic examination of Giemsa stained smears, ethidium bromide detection on amplified B. microti DNA and Southern blotting and ECL detection on amplified B. microti DNA were 3.2 %, 17.5 % and 47.6 %, respective. Sensitivity of the last detection system was high, being able to detect 13 molecules per 5 ul DNA solution. Sequence alignment of ss-rDNA fragments of our specimen and B. microti isolates from the United States, South Africa and Kobe, Japan revealed that Taiwan and Kobe isolates had a homology of 99 %, whereas homology between Taiwan and United States and South Africa were 96 %. Indicating that Taiwan and Japan B. microti isolates were more closely related.
