PCR products were separated by agarose gel electrophoresis and directly assigned the genotypes. Gene frequencies, power of discrimination (PD) and power of exclusion (PE) of the seven Alu insertion loci were established. There was no mutation observed in this family study. The power of discrimination were 0.1606 (HS 3.23), 0.6181 (HS 4.32), 0.6242 (PLAT), 0.5670 (ACE), 0.2198 (APO) and 0.6203 (B65), and the combined discrimination power was 0.985. Because of their stability, Alu insertions could help when suspect mutation was observed and couldn’t be confirmed in parentage testing. Besides, two primer multiplex PCR were tested and correct products were found on the combination of TPA and ACE loci. After the Alu insertions were sequenced, direct repeats and flanking sequences could be obtained. The results could be used to design distinctive probes of long and short products, which could be used in blotting analysis. Because of the dimorphism of Alu insertions, they might be able to be used in the system of microarray in the future. This study demonstrates the “master gene” model of its evolution. 李俊億 2001 學位論文 ; thesis 107 zh-TW
