After collecting compost samples from various composting fields, we obtained bacterial identification and classification according to Biolog MicroStation System. The results showed that the bacterial flora at 30℃ mainly comprised Staphylococcus spp., Corynebacterium spp., Acinetobacter spp., and CDC group E. At 40℃, the bacterial flora mainly comprised Corynebacterium spp., Acinetobacter spp., CDC group, and Bacillus spp.. At 50℃, the bacterial flora mainly comprised Bacillus spp. and Turicella otitidis. As the growth temperature increased to 60℃, the bacteria flora mainly comprised Bacillus spp.. Currently, by phenotypic methods of classification, bacteriologists are yet unable to distinguish and classify many different bacteria in the genus of Bacillus. Nonetheless, owing to the recent application of 16S rDNA analysis, a few new genera and species have been spun out from the genus of Bacillus. Therefore, we synthesized a pair of specific primers according to the sequence of the highly conserved region of 16S rDNA, and amplified the 16S rDNA of the thermophilic bacterial isolates by polymerase chain reaction (PCR). The resultant DNA fragments were sequenced, and the sequences were compared with that of the 16S rDNA listed in GenBank. Based on the 90% similarity in 16S rDNA sequences, five and one thermophilic bacterial isolates were categorized respectively into the genus Bacillus and the genus of Paenibacillus.
