CⅡ is an abundant molecule in articular cartilage and has been implicated as a possible autoantigen for RA. Oral administration of CⅡ has been show to suppress arthritis in several animal models, and several clinical trials have also shown positive results. Although there are many studies on CⅡ from bovine cartilage and chicken sternal against RA, little is known about the properties of jellyfish collagen in general. We obtained collagen substances in native form from Rhopilema sp., Stomolophus meleagris and commercially processed jellyfish products by pepsin limited digestion, solid NaCl fractions, and CM-Toyopearl chromatography. The yield was 55-64% and 23% of dried weight from edible and inedible jellyfish species, respectively. The chain composition of collagen derived from Rhopilema sp. seemed to be (1)3. The results of peptide mapping, amino acid composition, and carbohydrate content of jellyfish collagen were different from the C I, CII & CV of vertebrate animals. On the other hand, collagen extrected from Rhopilema sp. was low in proline, hydroxyproline & high in aspartic acid, hydroxylysine, and carbohydrate content, which was in line with other marine invertebrate creatures. In the present study, collagen materials derived from both Rhopilema sp. and Stomolophus meleagris were efficacious in inducing oral tolerance on collagen induced arthritis (CIA) at different dosages in rats, the CIA symptoms were assessed by visual examination, caliper measurement, and histological evaluation. Jellyfish collagen showed a comparable result to bovine CⅡ in suppressing CIA, but failed to delay the onset of the disease. Collagen from Rhopilema sp. was not able to induce arthritis on rats, it’s lacked for immunogenity.
