One and seven month old male senescence accelerated mice ( SAMP8 ) were divided into four groups : 20 % casein diet group ( control group ) and 20 % casein diet supplemented with low ( 0.1 % ), medium ( 0.25 % ) and high ( 0.5 % ) concentrations of Polygonum multiflorum Thunb. extracted by 50 % ethanol for 18 weeks. Body weight, food intake, open field activity test, single-trial passive avoidance and active shuttle avoidance test were performed. The neurotransmitter, malondialdehyde ( MDA ), vitamin C and total thiol of brain ( hippocampus, striatum and cortex ), heart and liver were analyzed after sacrifice, serum lipids were also evaluated. The results showed that there were no significant differences in the body weight, food intake, and locomotion within four groups in both 1- and 7- month old mice, but 1 month old groups had higher feed efficiency. Single-trial passive avoidance test showed 1 and 7 month old mice fed with 0.25 % of Polygonum multiflorum Thunb. extract had significantly better learning and memory. The groups fed with different concentrations of Polygonum multiflorum Thunb. extracts in both age had better active shuttle avoidance response, and 1 month old mice were better than 7 month old mice. In 1 month old mice, the group fed with 0.1 % of Polygonum multiflorum Thunb. extracts had higher norepinephrine、dopamine、dihydroxyphenylacetic acid and homovanillic concentrations in striatum, and dihydroxyphenylacetic acid concentration in cortex. 0.1 % of Polygonum multiflorum Thunb. extracts also increase 5-hydroxyindoleacetic acid concentration of cortex in both 1 and 7 month old mice. The MDA concentrations of hippocampus and cortex in 0.25 % of Polygonum multiflorum Thunb. extracts groups were significantly lower, but there were no differences among 7 month old groups. 7 month old mice fed with 0.5 % of Polygonum multiflorum Thunb. had higher vitamin C concentrations in liver, and the 1 month old mice had higher total thiol in liver and lower serum total cholesterol and triglyceride. In summary, we conclude that the supplement of Polygonum multiflorum Thunb. extracts could improve the abilities of learning and memory, increase brain neurotransmitter concentrations, promote the antioxidative system and lowering the oxidative injury, it may be effective to the aging.
