In general, these wounds are repaired with split-skin grafts. Cultured human epithelial autografts (CEA) are alternatives for wound closure. Injured tissue also can be repaired by covering with a non-medicated or medicated wound dressing. In addition wound dressing can be used for skin lesion such as, burn injury, surgical wounds and chronic ulcer. The Chinese herb Bletilla striata has been used to treatment peptic ulcer and cutaneous wound in clinics. In the present study, we first evaluated the mutagenic activity of Bletilla striata. As a resutt, Bletilla striata did not have mutagenic activities in the Ames test using Samonella typhimurium strains TA97, TA98, TA100, and TA102, with and without metabolic activation. Electrophretic analysis of protein on SDS-page was used to detect protein content and protein contain below to 0.005% in extract solution of Bletilla striata. Cytotoxicity test was arranged to assess the Bletilla striata cytotoxicity to human keratinocytes. Results of MTT assay showed the cell counts increased with low concentration of Bletilla striata solution. In vivo tests, we evaluated the effects of Bletilla striata gel (10% , 20%) on repairing the dermal damages of guinea pigs. Results showed Bletilla striata gel (10% , 20%) could promote keratinocytes migration, angiogenesis, and granulation tissue formation. In order to control the drug efficiency, we used a genipin-fixed gelatin to encapuslate the with Bletilla striata gel. Different bloom numbers of gelatin (300, 225, 75) were used to prepare wound dressings with different degradation rates. Three types of wound dressings were then applied on the full-thickness dorsal skin defect of Sprague-Dawley rat. At postoperative day 10 and 20, histological results showed an enhancement of wound healing in all the genipin-fixed gelatin matrices, especially the one with bloom number 300.
