Levels of gastric ulcer index(UI)、gastric nonprotein sulfhydryl groups(GNPSH)、liver nonprotein sulfhydryl groups(LNPSH)、gastric lipid peroxidation (GLPO)and plasma lipid peroxidation(PLPO)were examined after WIRS stress over 2h or 4h period among five groups with pretreatment of five different doses(25, 50, 100, 250 and 500 mg/kg B.W.)of Chai-hu-gui-zhi-tang extract. Besides, the antioxidant effects of the medicine on normal Wistar rats for 2h or 4h administration and 3 weeks administration were evaluated in this study to assess Chai-hu-gui-zhi-tang as an alternative drug for the clinical prevention of gastric hemorrhage. With 2h of WIRS induction, levels of gastric and plasma lipid peroxidation highly significantly incresaes with the development of gastric lesion(P < 0.01), while levels of gastric nonprotein sulfhydryl groups significantly decreased(P < 0.05). With 4h of WIRS stress, levels of gastric lipid peroxidation highly significantly incresaes with the development of gastric lesion(P < 0.01), while levels of gastric nonprotein sulfhydryl groups highly significantly decreased(P < 0.01)with significantly decreased of liver nonprotein sulfhydryl groups concentration(P < 0.05). Pretreatment of the medicine highly significantly attenuated the development of gastric lesion induced by 2h and 4h of WIRS stress among 25, 50, 100, 250 and 500 mg/kg B.W. groups(P < 0.01). The decrease of gastric nonprotein sulfhydryl groups concentration induced by 2h and 4h of WIRS stress was significantly or highly significantly attenuated by pretreatment of the medicine among 100, 250 and 500 mg/kg B.W. groups(P < 0.05 or P < 0.01). The decrease of liver nonprotein sulfhydryl groups concentration induced by 4h of WIRS stress was significantly attenuated by pretreatment of the medicine among 250 mg/kg B.W. group (P < 0.05). However, the increase of gastric lipid peroxidation concentration induced by 2h and 4h of WIRS stress could not be attenuated by pretreatment of the medicine among all groups(P > 0.05). Furthermore, pretreatment of the medicine would significantly increased the levels of gastric lipid peroxidation with 2h and 4h of WIRS among 100 mg/kg B.W. group(P < 0.05). Pretreatment of the medicine would also highly significantly increased the levels of plasma lipid peroxidation with 4h of WIRS stress among 25, 50, 100, 250 and 500 mg/kg B.W. groups(P < 0.01). In normal rats, adminstration of the medicine for 2h or 4h promoted significantly or highly significantly increase of gastric nonprotein sulfhydryl groups concentration and liver nonprotein sulfhydryl groups concentration, and caused significantly or highly significantly decrease of plasma lipid peroxidation concentration among 25, 50, 100, 250 and 500 mg/kg B.W. groups(P < 0.05 or P < 0.01). Administration of the medicine for 2h caused highly significantly increase of the levels of gastric lipid peroxidation and plasma lipid peroxidation among 100 mg/kg B.W. group(P < 0.01). Long time administration of the medicine at a dose of 250 mg/kg B.W. daily for 3 weeks did not cause significantly changes to the levels of gastric nonprotein sulfhydryl groups、liver nonprotein sulfhydryl groups、gastric lipid peroxidation and plasma lipid peroxidation among normal aged group(Ten months old)(P > 0.05). In normal young group(Ten weeks old), long time administration of the medicine at a dose of 250 mg/kg B.W. daily for 3 weeks caused significantly increase of liver nonprotein sulfhydryl groups concentration(P < 0.05). In conclusion: Chai-hu-gui-zhi-tang extract could prevent the development of acute gastric lesions and the decrease of gastric nonprotein sulfhydryl groups concentration induced by WIRS. In normal rats, short time administration of the medicine would promote the decrease of plasma lipid peroxidation concentration and the increase of gastric nonprotein sulfhydryl groups concentration and liver nonprotein sulfhydryl groups concentration. In normal young rats, long time administration of the medicine would promote the increase of liver nonprotein sulfhydryl groups concentration. Therefore, Chai-hu-gui-zhi-tang extract is also an effective antioxidant.
