In our experiment, we not only use Caco-2 cell monolayer as an experimental model to imitate real intestinal epithelial cell, but also use it to study the effects of the probiotic Lactobacillus. GG adhered on Caco-2 cell monolayer during antibiotics and flavonoids transport. At first, we cultured Caco-2 cell and set up the Caco-2 cell bank, and utilized it as an adhesive and transported experimental model. The optimal adhesive conditions of the 18 hours incubated L. GG was at the 0.490 OD600 absorption and 2 hours incubated conditions after added to the Caco-2 cell monolayer. In the flavonoids and antibiotics transport experiments, 60μM Quercetin had a better apparent permeability (Papp) and could accumulate more amount in blood, but after L. GG adhered to the Caco-2 cell monolayer, the transport of Quercetins were all decreased. In transport test of Chrysin, it can transport through the intestinal model, but its total absorption was limited by higher Bl-Ap transport. After L. GG adhered, Chrysin could increase its Papp value only under 60μM. From these two results, we supposed that L. GG adhered on Caco-2 monolayer to form a barrier to block the transport of the two substances. In antibiotics transport assay, hardly transported Gentamicin and well-transported Erythromycin could increase their Papp value after L. GG adhered. But there was no apparent effect on transport of Neomycin.
