Antracence-2, 3-dicarboxyaldehyde (ADA) and 5-carboxy tetramethyl rhodamine, succinimidyl ester (5-TAMRA, SE) were used for precolumn derivatization of the non-fluorescence drug. The relative standard deviations value on peak area and migration time were 9.44%, 0.88% for ADA-VGB derivative and 9.48%,0.86% for 5-TAMRA-VGB derivative, respectively. The concentration limit of detection (S/N=3) in standard solution was calculated to be 0.024 mM for VGB. Coupled with a simple cleanup procedure, the method was successfully applied to the analysis of Vigabatrin in human plasma. Recoveries of spiked 100mM and 8mM Vigabatrin in plasma were 100.25% and 95.15 %. The assay was linear between the concentration range 1.5-200mm with a correlation coefficient (r) of 0.9958, and the limit of detection (S/N=3) of Vigabatrin in plasma was calculated to be 0.13mm . The inter-day and intra-day reproducibilities and accuracies of both spiked 100mM and 8mM in plasma were less than 14.29% and 4.88%, respectively. Yu-Chen Chang 張玉珍 2003 學位論文 ; thesis 79 zh-TW
