It has been widely applied in the industries of foods, cosmetics, biomaterials, etc. Its quantification is mainly based on the hydroxyproline assay of samples by chemical chromatic analysis. Since the method is somewhat tedious and non-specific to non-vertebrate samples, the current study focused on establishing an easy, sensitive, and accurate method for the quantification of collagen by using capillary electrophoresis. The specificity and reproducibility were first characterized after identifying the optimal assay system for collagen. Various animal tissues were analyzed by the traditional hydroxyproline assay, hydroxyproline quantification by capillary electrophoresis, and direct collagen quantification by capillary electrophoresis. Our results demonstrated that no significant difference appeared on the data measured by the above variant methods. This indicates that the quantification method developed by using capillary electrophoresis is feasible and accurate. One of the advantages is that our method vastly reduced the timing for waiting and working from about 7 days to less than 3 days which include the time for sample processing. Lynn L. H.
