Dengue virus type 2 ( DEN2 ) has a single-stranded RNA genome ,containing 10,723 nucleotides . DEN2 RNA encodes a polyprotein precursor , which is processed into three structural proteins（C : core protein, M : membrane-associated protein , E : envelope protein） and at least seven nonstructural proteins NS1,NS2A, NS2B,NS3, NS4A, NS4B,NS5 , which are expressed in infected cell .The nonstructural protein NS3 possesses multiple enzymatic activities and is very important for viral replication. The N-terminus of NS3 protein possesses the serine protease activity and the C-terminus of NS3 protein contains the nucleoside triphosphatase (NTPase), helicase and RNA triphosphatase activities. In the study , we have expressed the NS3-C protein in Pichia pastoris for the ATPase , RNA triphosphatase activities assay . So far, only ATPase activity was observed.
