In the process of BBP production, experiment was designed through Response Surface Methodology (RSM), and the optimal medium was taken from the effects of constituents on enzyme production. It was found that the corresponding concentrations were - shrimp shell powder 0.72%, soy powder 0.6%, sucrose 0.19% and yeast extract 0.68%. The theoretical concentration of enzyme production is 281.14 unit/mL, which coincides with the shaker-flask cultivation value 280.72 unit/mL. It reveals the reliability of RSM mode. Based on these results, experiment was extended to 5-L stirred tank reactor (STR), and 30-L airlift bioreactor. In 5-L STR at an aeration rate 0.6 vvm, temperature 26 �aC and agitation rate 150 rpm, the maximum enzyme production was found 238.77 unit/mL. Whereas in 30-L airlift bioreactor high yield 283.84 unit/mL of protease was obtained at aeration rate of 0.9 vvm and temperature 26 �aC for three days cultivation.
