Description
Summary:碩士 === 高雄醫學大學 === 醫學研究所碩士班 === 92 === Some Chinese medicinal herbs have been used as health food. In order to investigate the biological activities in 17 kinds of Chinese medicinal herbs including Foeniculum vulgare Mill, Polygonum cuspidatum Sieb. et Zucc., and Bupleurum chinense D.C., the following experiments were carried out. In the study of anticancer activity, it was used XTT to screen and determine the IC50 values for human hepatoma, leukemia and skin cancer cell lines. Further, we used DNA fragmentation and flow cytometry to demonstrate the activity of inducing apoptosis. In the study of anti-HBV activity, we used Real-time PCR to quantify the amount of HBV DNA and determined the secretion of HBsAg and HBeAg by ELISA kit. Psoralen is a pure compound of F. vulgare. It possesses antileukemic activity and IC50 ranged from 15.66±0.42 to 23.16±2.43mg/ml. The IC50 of emodin against skin cancer cells BCC-1/KMC was 5.27±0.79mg/ml. In the DNA fragmentation analysis after treatment with emodin, we found the ladder mode of DNA via electrophoresis. Furthermore, in flow cytometry analysis, results showed that the area of subG1 peak was increasing, and the cell cycle was arrested at G2/M phase. Following these results, we concluded that the emodin effectively inhibited cell growth and induced apoptosis in human skin cancer cells. The results of anti-hepatitis B virus activities revealed that the amount of extracellular HBV DNA was reduced by sho-saiko-to, water extract and saikosaponins of B. chinense, whereas the water extract and ethanolic extract of P. cuspidatum suppressed the secretion of HBeAg and the amount of extracellular HBV DNA.
